Quantitative gravimetric fractionation of brain tissue.
نویسنده
چکیده
With the procedure which is described, brain tissue is separated into six fractions, the sum of whose weights equals the dry weight of the tissue. Three of these primary fractions are further separated into subfractions. Recovery is quantitative at each step. The procedure, as applied to rat brain, gave consistent results; duplicate analyses of the two halves of brains, divided as equally as possible along the anteropostenor line, agreed closely, and the variation from rat to rat in the percentages of the various fractions in dry tissue was small, as illustrated by the following results: main lipid fraction, 40.65 ± 1.03 (S.D.) %; protein residue, 44.45 ± 0.87%; water-soluble fraction from Folch purification, 5.94 ± 0.23%; crude “strandin” fraction, 1.25 ± 0.05%. The average percentage of dry in fresh tissue was 24.87 ± 0.41. The method includes procedures for the separation of a fraction, obtained by treatment with acidified chloroform-methanol, into four well-defined fractions, of which one is characterized by extreme surface activity in aqueous solution. A small amount of lipid is so firmly bound in brain tissue as to resist extraction during exhaustive treatment with chloroform-methanol (2:1), acidifed chloroform-methanol, and acetone. Significant radioactivity was present in most of the fractions from the brains of rats which had received a peritoneal injection of acetate-C” 5-15 months previously during the period of early development.

Quantitative Comparison of Analytical solution and Finite Element Method for investigation of Near-Infrared Light Propagation in Brain Tissue Model
Introduction: Functional Near-Infrared Spectroscopy (fNIRS) is an imaging method in which light source and detector are installed on the head; consequently, re-emission of light from human skin contains information about cerebral hemodynamic alteration. The spatial probability distribution profile of photons penetrating tissue at a source spot, scattering into the tissue, and being released at ...
متن کاملQuantitative measurements of retinal edema by specific gravity determinations.
We have quantified retinal edema in rat by specific gravity measurements in vitro. Specific gravity corresponds to tissue water content, which is increased in retinal edema. The gravimetric analysis of retinal edema corresponds well with light and transmission electron microscopy and radiolabelled albumen studies. Specific gravity measurements are the standard method to measure brain edema and ...
متن کاملTumour radiobiology beyond fractionation
Historically it has been shown repeatedly that single high doses of radiation do not allow a therapeutic differential between tumor and critical normal tissues but dose fractionation does. The purpose of conventional dose fractionation is to increase dose to the tumor while preserving normal tissue function. Tumors are generally irradiated with 2Gy dose per fraction delivered daily to a more or...
متن کاملQuantitative Comparison of SPM, FSL, and Brainsuite for Brain MR Image Segmentation
Background: Accurate brain tissue segmentation from magnetic resonance (MR) images is an important step in analysis of cerebral images. There are software packages which are used for brain segmentation. These packages usually contain a set of skull stripping, intensity non-uniformity (bias) correction and segmentation routines. Thus, assessment of the quality of the segmented gray matter (GM), ...
متن کاملPathological Assessment of Brain White Matter in Relapsing-Remitting MS Patients using Quantitative Magnetization Transfer Imaging
Introduction: Multiple sclerosis (MS) is characterized by lesions in the white matter (WM) of the central nervous system. Magnetic resonance imaging is the most specific and sensitive method for diagnosis of multiple sclerosis. However, the ability of conventional MRI to show histopathologic heterogeneity of MS lesions is insufficient. Quantitative magnetization transfer imaging (qMTI) is a rel...
